Cerkiew znajduje się przy ulicy Ostrobramskiej. Została ona założona przez prawosławne bractwo Św. Trójcy w 1597 roku. Świątynia ta szybko jednak spłonęła, więc w 1638 roku wzniesiono murowaną już świątynię na planie krzyża łacińskiego. Po kolejnym pożarze w roku 1749 powstał rokokowy ikonostas. Wnętrza cerkwi zdobią obrazy rosyjskiego malarza Iwana Trutniewa. W podziemiach natomiast przechowywane są relikwie świętych: Jana, Antoniusza i Eustachego. Byli to dworzanie ruskiej księżniczki Julianny, których w XIV wieku w Wilnie zamordowali pogańscy wówczas Litwini.

The Orthodox Church of the Holy Spirit was on the way from my hotel to whatever sight I was seeing in Vilnius, and I decided to drop in. What a beautiful place of worship. No tickets are necessary. It's one of the many churches in Europe that might not be on a lot of "must see" lists, but it will absolutely take your breath away!

Przepiękna cerkiew do której weszliśmy przez przypadek wracając z Ostrej Bramy. Tu kiedyś było więzienie w którym Mickiewicz napisał Dziady Jest tu celach Konrada. Ale najbardziej zafascynowało nas wielki przeszklony sarkofag po środku świątyni w którym leżały mumie trzech świętych z 14 wieku. Podobno ich mumie powstały w sposób naturalny, bez udziału ludzi.Ich ciała są okryte natomiast wystają nogi w białych skarpetach. Młoda kobieta przez dłuższy czas stała z swieczkami przy nich i płakała.
